I don't have a Gen IV game yet (DPP). I've seen the movesets are very different in DPP than previous versions.

If I have a pokemon, hypno for instance, that is already level 100 from Emerald, how do I get the level up moves from DPP on him?

The answer is quite simple. When you get Diamond/Pearl/Platinum, have beaten the game and obtained the national dex and unlocked the pal park, thus being able to transfer your pokemon from Emerald/any of the other Geneartion III games to Diamond/Pearl/Platinum. Take the said Hypno to the move tutor in Pastoria City, and for a Heart Scale we will teach the Hypno any of the moves he could have learned if he was in the fourth gen.

Just to clarify: Do you have to play through the entire game before you can trade in any of your old favorites?

I believe so, but all I know for certain is you need the national dex.
